I like to use a small amount on the end of the spark plug boot. this allows the plug wire to be removed easier later.

It's not strictly necessary, as the plugs will still function without it. But it's a good idea, as it helps prevent corrosion as well as allowing the wire to be removed more easily as Nick already said.
