Basic interrogation, like a lie detector, works on the theory that people respond pretty much the same to similar stimulus. Being confronted with inconsistencies that make the witness ether lie or confess to something makes them uncomfortable, and the biophysical reaction to that discomfort can be observed, either by a machine or a trained observer. Since a sociopath has few, if any social inhibitions, it's hard to detect any discomfort when he lies.

I've always heard, (and believed), that pathological liars actually believe their lies at some point; i.e. OJ Simpson on some twisted level believes that he is innocent.
