pull a plug then try to start the car. shut off car. rinse and repeat. if it is a spark issue you may be able to notice a different idle. Another option would be to get/make a spare wire and swap till it is running smooth...

pulling wires one at a time is the fastest way. the dead hole will show up right away. so what if you get a tiny tingle, it ain't gonna kill ya. bunch of wimps. MSD on the other hand might. another way is to loosen each fuel line bolt or nut one at a time up on the fuel distributor. minor fuel wetness, not much, and you only need to do it for a second or two. keep a rag handy. 3.2s are easiest. just unplug one injector at a time.

Safety question: Is it safe to pull and plug these in with the motor running?

you will only do it once!:D

Tiny tingle? :D

BTDT. Not recommended if you have a pace maker.
It's especially good when you lean in deep to do #3 & 6 and touch ground

John: Loosen the fuel line at the FD?
Is that for when you suspect air in the line?? Or??

no. it kills the cylinder just like pulling a plug wire.

Follow Up to my original post.

So after checking, plugs, wires, vacuum lines, distributor, etc. the problem turned out to be a faulty Oxygen sensor which was causing the motor to run way too lean. I never looked at this since my sensor probably has 500 miles on it. I replaced the old one it last summer. Thanks to Dan Jacobs for solving the mystery.
