I have a 2006 Mini Cooper R50 5 speed transmission and when I was going to change the trans fluid, I saw a note on the transmission that states No Oil change required.
Should I change it or not?

Yes, that is lifetime fluid. However, MINI does not define lifetime. I would suggest replacing it.
