Best comedy duo ever, considered a poll, but knew I'd forget someone
Best comedic pair ever, considering joint and independent work.
I go with Mel Brooks and Carl Reiner. Laurel and Hardy, Abbott and Costello, Tim Conway and Harvey Korman, Gleason and Carney. I would get to thinking about it, and was sure I would leave out somebody, hence did not do a poll.

Given the OP's criteria, joint and independent work, I'll go with his choices--Brooks & Reiner and Conway & Korman.
Though the others mentioned are funny, their humor was scripted. Both Brooks/Reiner and Conway/Korman would typically begin with a scripted routine and then take off on their own hilarious tangent. That, to me, is the true source of the best humor.
