I imagine it could be done either way, but it is definitely easier to access the rear glass with the parcel shelf removed.
Unlike the dash which has fasteners that are impossible to access with the windshield installed, the parcel shelf slides into position at the base of the window and all the fasteners are located in the rear seat area.
