Very possible that your tooth gap is not aligned with TDC. So you have to take into account the missing tooth offset.

Honestly, It doesn't matter what the offset is as long as your timing firing with a light matches the indicator on the crank pulley. Meaning. zero deg on ECU matches 0 deg on pulley. Don't make this too complicated.

I admit I have never heard of Emu (other than a bird). Is this sequential with a cam phasing trigger signal? It is possible that you are 180 out and trying to fire #1 at #4. That is called wasted spark and will not run. Add 180 to the offset or rephase your cam signal.
