I used to tune the night of the gig - sure, transport can make a big difference even playing for more than a few hours at a constant ambient temp will. Some songs you can get by without and if you do some serious heavy metal you may have them muffled to sound like your hitting a plastic garbage can but for music I used to play which was big band, swing and jazz (both traditional and modern) than a nice melodic tom set up with tuned double heads and no muffle actually sound pretty darn sweet.
