All good ideas so far.

If you're not used to all-day riding, you can expect some leg cramps and other aches and pains. Some warm-up trips or being conscientious about your stretches for a few weeks can help.

The greatest space-age miracle creation is: long-hair sheep skin. Esp for ventilation. Buy one at Ikea for $20 and just sit on it.

Having spongy soles on your boots helps too and often isn't thought of. Also tall to keep the breeze off your legs. Helps in the way ear plugs help.

Best to have non-flapping clothes on a long trip.

In addition to all the self-evident and/or well-known ergo fixes, you can raise the fork stanchions two inches and install either the original or the "comfort" clip-ons over the upper clamp.

Isn't somebody going to mention the Leatt-Brace neck gizmo that was all the lemming-rage last year, from South Africa with a Nevada liability-avoidance corporate registration and some other corporate aspects that seemed to me to have a bad smell. Might be good but I'd like to see some test results if anybody can point to them.
