I like them both for different reasons. South Beach for the nightlife & the architecture. Key West for its laid back lifestyle.
Also, if you like snorkeling/diving/kayaking, I'd suggest a stop in Key Largo. There's the Pennekamp State park.
Either way, you've got plenty of sandy beaches if you want to do absolutely nothing & just relax.
BTW, it's about a 4 or 5 hour drive from Miami depending on traffic.
