Perhaps you should invest in a spark tester, might save your life, lol.

On my car, I can unplug #2 or #4 while it is running, and it makes no difference, but I guarantee both are firing.

Try laying the spark plug on the valve cover, and trying to start it, see if you get spark on the plug. I'm presuming you cleaned the connector inside the boot where it attaches to the plug, I guess that could be it as well.
