I'd be most worried about the caldera underneath Yellowstone or a near Earth object. But then I also worry about the number of people who don't believe those things will ever happen.

Religious differences will likely lead to a nuclear exchange between two or more nations/powers leading to the eradication of millions, with billions more dying off slowly in a polluted, radioactive world with little capacity to produce food and agriculture in the years following.
This is the most likely scenario as I see it. Frankly I'd be shocked if the human race survives another 500 years. Honestly, I think this scenario will happen in the next 100. Quite possibly in our generation. Our restraint has historically never kept pace with our technology and we still (as a species) are inclined to cling to silly ancient mysticism, religious superstition and tribal-based xenophobia rather than trying to see ourselves as part of a collective race of inhabitants on one very small planet capable of supporting life in a vast ocean of barren emptiness.

I'd guess that a superbug would wipe out a good deal of the population, a la the black plague. Mass hysteria ignites, and a slow crumble from there.
Or a gigantic asteroid will erase any memory of us being here.

We'd probably get some advanced notice of a NEO hit. Amazing how so many people think the movie Deep Impact is science fiction. It's very much science non-fiction. Imagine if the comet that hit Tunguska had actually hit the mid-Atlantic today. And that was only around 100 yrs. ago. There have been countless others and will be countless more. What are the odds we could detect and deflect or destroy the objects that caused Tunguska or Meteor Crater in AZ if they were to happen today? Whatever does us in, I hope I'm gone in the initial hit. I don't think I want to stick around to see who dies last when the crops disappear after a year of no sunlight.
